Данная работа является частью проекта по созданию "ловушек" вирусных частиц. Создание подобных "ловушек" происходит за счет ковалентной иммобилизации на поверхности нано- или микрочастиц на основе полимолочной кислоты (ПМК) рекомбинантного рецептора, специфического для данного вируса. Предполагается, что полученные конъюгаты белок-частица будут способны к связыванию с вирионами, с последующим поглощением клетками иммунной системы организма. Целью работы являлось изучение влияния полимерных частиц на иммуногенность модельных белков. Произведены иммунизация и забор крови 80 мышей, разделенных на 2 группы: "Контроль" и "Опыт", 4 раза с интервалами 2 недели. На каждом этапе иммунизации содержание антител в Контрольной группе мышей значимо выше, чем в Опытной группе (p<0,004).
We investigated the immunogenic properties of proteins associated with nanoparticles. A process for producing nanoparticles by the polymerization of lactic acid and an optimal method of surface activation were described. Activated nanoparticles used for covalent binding of beta-2 microglobulin protein with green fluorescent protein. It is shown that nanoparticles able to bind 3.03 micrograms per 1 mg of the polymer. According to confocal microscopy and electrophoresis the protein firmly adsorbed on the surface of the granules. Mice line F1 (CBA x C57BL) were intraperitoneally immunized by nanoparticles modified fusion protein and the equivalent of a mixture of unmodified nanoparticles with fusion protein. Blood was taken at 2 weeks after three times intraperitoneal immunization. Antibody concentration to protein was determined in mouse serum using enzyme immunoassay. Experimental and control groups made up to 40 animals. The validity of the results was evaluated using the Mann-Whitney test. It is shown that the average concentration of antibodies in the control group is 1.8 times greater than that in the experimental group. The difference was significant. We discuss the significance of the results to create traps capable of binding the virus particles in the blood and provide an immune response.
