В работе исследуются условия кристаллизации рекомбинантного цито-плазматического сигма-1 рецептора человека. В ходе работы ген цитоплаз-матического домена сигма-1 рецептора человека был клонирован в бактери-ях E.coli, создана система синтеза и очистки рекомбинантного цитоплазмати-ческого домена сигма-1 рецептора в составе гибридного белка в комплексе с мальтоза-связывающим белком. Методом бесцветного нативного электрофо-реза показано, что раствор исследуемого белка представляет собой полидис-персную смесь олигомеров и мономеров. Разработаны две стратегии реше-ния проблемы полидисперсности: применение детергентов и недетергентных добавок и получение мутантных форм цитоплазматического домена с точеч-ной мутацией в сайте олигомеризации GXXXG.
In this thesis the crystallization screening of the CTD of recombinant hu-man sigma-1 receptor CTD is carried out. The gene of human sigma-1 receptor CTD has been cloned and the CTD fused to MBP by 4 alanine amino acid se-quence has been overexpressed in E. coli bacteria. The downstream process of protein extraction and purification has been optimized. The result of CN-PAGE analysis showed that the protein of interest exists in a solution as a mixture of monomeric and homooligomeric forms. To address this issue, we undertook the studies to convert the CTD of human sigma-1 receptor to the homogenous state. These studies have shown that using different detergents and crystallization addi-tives may be one of the ways to solve this problem. The second way is to design mutant forms of sigma-1 receptor CTD by replacing one of the amino acids in GXXXG oligomerization site.
