Гигантские бактериофаги phiKZ, заражающие бактерии Pseudomonas aeruginosa, осуществляют транскрипцию своего генома при помощи исключительно своих двух ДНК-зависимых РНК-полимераз: вирионной и невирионной, не привлекая РНКП клетки-хозяина. Было показано, что в ходе инфекции бактериальных клеток фагом phiKZ во второй половине инфекции внутри клетки образуется сферическая структура внутрь которой упаковывается ДНК (псевдо-ядро). Положение обеих фаговых РНКП относительно данной структуры вероятно связано с регуляцией транскрипции фагового генома. Оценка положения обеих фаговых РНКП была проведена в ходе работы с использованием метода флуоресцентной микроскопии.
Giant bacteriophages phiKZ, which infect Pseudomonas aeruginosa bacteria, implement their genome’s transcription by their own two DNA-dependent RNApolymerases: virion RNAP and non-virion RNAP, without RNAPs of the host cell. It has been shown that during infection of bacterial cells with the phiKZ phage a protein spherical structure is being formed, in which the DNA is being packed to (psedonucleus). The location of the both phage RNAPs relatively to this structure is possibly related with the phage genome transcription regulation. An assessment of both of the RNAPs’ location has been performed in this work using the fluorescent micros copy method.
