В данной работе была изучена репродуктивная активность реассортантных штаммов вируса гриппа, содержащих нуклеопротеин (NP) от вирусов различных лет выделения. В ходе парного сравнения вакцинных штаммов живой гриппозной вакцины (ЖГВ), содержащих NP от устаревшего штамма - донора аттенуации (формула генома 6:2) и от современного вируса H1N1 или H7N9 (формула генома 5:3) выявлено разнонаправленное влияние NP гена на ростовые характеристики ЖГВ. Для вируса подтипа H1N1 было свойственно снижение активности репродукции 5:3 реассортанта при оптимальной температуре инкубации, как в развивающихся куриных эмбрионах (РКЭ), так и в культуре клеток MDCK, тогда как у вируса H7N9 наблюдалось усиление ростовых характеристик при замене NP гена на более современный. Включение современного NP в геном донора аттенуации драматически влияло на репродукцию вируса в РКЭ при пониженной температуре инкубации, однако этот эффект практически нивелировался при включении в реассортантный штамм поверхностных антигенов HA и NA от соответствующих эпидемических вирусов. Также показано, что реассортантные штаммы ЖГВ с формулой генома 5:3 сохраняют ts/ca фенотипы в РКЭ, присущие классическим реассортантным штаммам с формулой генома 6:2.
Were studied the reproductive activity of reassortant influenza virus strains containing nucleoprotein (NP) from viruses isolated in different years. A side-by-side comparison of live attenuated influenza vaccine (LAIV) strains containing NP from old strain - master donor virus (genome composition 6:2) and currently circulating H1N1 or H7N9 viruses (genome composition 5:3) revealed multidirectional effect of NP gene on growth characteristics of LAIV. Infectious titers of 5:3 reassortant of H1N1 subtype were significantly lower in eggs and MDCK cells, compared to 6:2 reassortant. In contrast, H7N9 reassortant virus became more infectious when the old NP gene was replaced with the corresponding gene of wild-type influenza virus. The inclusion of wild-type NP in genome of the master donor virus dramatically affected the reproduction of the virus at low temperature in CE. However, this effect almost disappeared when corresponding wild-type surface antigens, HA and NA, were added to the reassortant viruses. Importantly, the LAIV reassortant strains with 5:3 genome composition had the same ts/ca phenotypes as the classical reassortant strains with 6:2 genome composition.
