Изучение стволовых клеток эндометрия (СКЭ) является потенциально важной задачей для введения их в практическое применение. Целью работы являлось идентифицирование основных типов ионных каналов, экспрессирующихся в СКЭ. С помощью метода патч-кламп были идентифицированы кальций-активируемые калиевые каналы семейств SK и BK с различной чувствительностью к уровню внутриклеточного кальция. На уровне одиночных токов показан кальций и потенциал-зависимость каналов BK. Методом ОТ-ПЦР обнаружена экспрессия гена канало-формирующей субъединицы ВК канала, а также подтверждено присутствие ВК каналов в клеточной мембране с помощью иммунофлюоресцентного окрашивания.
Human mesenchymal endometrial stem cells (hMESCs) have several advantages over other adult stem cells isolated from different sources. For the potential use of hMESC in stem cell-based therapy, it is very important to characterize hMESCs on different levels. To date, the ionic mechanisms underlying physiological processes in hMESCs are unknown. Using the patch-clamp technique, several types of native potassium channels in hMESCs were identified on single current level. Moreover, the presence of two distinct types (SK and BK) of calcium-activated potassium channels was demonstrated. BK channels were activated in response to the increase of membrane potential (depolarization) and by the change in intracellular calcium concentration. It was shown that SK channels have higher calcium sensitivity comparing to BK. BK channel expression was confirmed by immunofluorescent staining and RT-PCR.
