Биомедицинское материаловедение и инженерия систем для персонализированной медицины занимает одну из лидирующих позиций в области науки и исследований современной медицины. Основное направление развития данного заключается в формировании полимерсомальных гибридных нано-и микроразмерных систем на основе биосовместимых-и(или) биодеградируемых материалов, инкасуляции в их объем биологических активных соединений (БАС), а также программирования их конечных свойств за счет внедрения модифицирующих агентов. Проблематика данного направления – это разработка эффективных методик синтеза исходных материалов, модифицирующих агентов и других компонентов, для создания гибридных систем – носителей с функцией контролируемого высвобождения биологически активных соединений (в том числе терапевтических веществ) в очаг воспаления мягких тканей и слоев эпидермиса. Решение существующей проблемы, позволит повысить терапевтический эффект различных групп БАС, действие которых направлено на превентивное действие к различным группам патогенных микроорганизмов, а также снижения времени регенерации. Используя методики формирования полимерсомальных гибридных систем методом коаксиального электрогидродинамического формования, было разработано лабораторно-экспериментальное устройство, с помощью которого были получены волокнистые и сферические ПГС с инкапсулированными антибактериальным полипептидным комплексом из катионного полипептида a,b – дефензина, полициклического пептида низина, а также веществом обладающим свойством ускорения процесса пролиферации клеток – декспантенолом. В результате проведенных исследований, используя сферические ПГС с инкапсулированным полипептидным комплексом была модифицирована поверхность хирургической нити. За счет модификации и проведенного флуометрического «Live/Dead» анализа на штаммах E.сoli и S.aureus была доказана эффективность использования данных структур, обладающих свойством контролируемого высвобождения и оказания превентивного антибактериального эффекта. Также, используя волокнистые ПГС с инкапсулированным декспантенолом, был создан перспективный материал для создания филамента хирургической нити, обладающий регенерирующим эффектом. Эффективность, была показана в результате in vitro эксперимента на клеточной культуре фибробластов, которые за счет влияния декспантенола увеличили скорость пролиферации.
Biomedical materials science and systems engineering for personalized medicine occupies one of the leading positions in the field of science and research in modern medicine. The main direction of this development is the formation of polymersomal hybrid nano-and micro-sized systems based on biocompatible and / or biodegradable materials, encapsulation of biological active compounds in their volume, as well as programming of their final properties due to the introduction of modifying agents. The problems of this direction are the development of effective methods for the synthesis of starting materials, modifying agents and other components to create hybrid carrier systems with the function of the controlled release of biologically active compounds (including therapeutic substances) into the focus of inflammation of soft tissues and epidermal layers. Solving the existing problem will increase the therapeutic effect of various BAC-groups, the action of which is aimed at the preventive effect on various groups of pathogenic microorganisms, as well as reducing the regeneration time. Using the methods of forming polymersomal hybrid systems by the method of coaxial electrohydrodynamic molding, a laboratory-experimental device was developed, with which fibrous and spherical BAC’s with an encapsulated antibacterial polypeptide complex from a cationic polypeptide a, b-defensin, a polycyclic nisin peptide, and also a substance possessing a property of accelerating the proliferation of cells - DE panthenol. As a result of the studies, the surface of the surgical thread was modified using spherical ASGs with an encapsulated polypeptide complex. Due to the modification and the fluometric "Live / Dead" analysis on strains of E.coli and S.aureus, the effectiveness of using these structures with the property of controlled release and the provision of a preventive antibacterial effect was proved. Also, using fibrous ASGs with encapsulated DE panthenol, a promising material was created for creating a filament of a surgical thread with a regenerating effect. Efficiency was shown as a result of an in vitro experiment on a cell culture of fibroblasts, which, due to the influence of DE panthenol, increased the proliferation rate.
