В работе были исследованы полиморфные варианты 5’-нетранслируемой области гена рецептора дофамина DRD1 и их влияние на экспрессию данного гена (мРНК, белок) в лимфоцитах периферической крови (ЛПК) у пациентов с расстройствами шизофренического спектра (РШС) и контрольной группы. Обнаружено снижение относительного уровня мРНК гена DRD1 у пациентов с РШС в сравнении с группой контроля, при этом значения количества белка DRD1 достоверно не различались между группами. В контрольной группе у носителей генетического варианта АА rs267410 (C/A) наблюдалась тенденция к снижению концентрации белка DRD1 в ЛПК, в сравнении с носителями вариантов СА и СС на границе статистической значимости (p=0.059). Было показано снижение относительного уровня экспрессии DRD1 у носителей минорного аллеля G в гомо- и гетерозиготном состоянии rs35916350(С/G) на границе статистической значимости (p=0.053) в контрольной группе. Обнаружено снижение концентрации белка DRD1 в ЛПК при терапии галоперидолом в течение 28 дней (p=0.001).
In this work, we investigated polymorphic variants of the 5'-untranslated region of the dopamine receptor gene (DRD1) and their influence on the expression of this gene (mRNA, protein) in peripheral blood lymphocytes (PBL) of patients with schizophrenia spectrum disorders (SSD) and control group. The mRNA level of DRD1 was significantly lower in patients with SSD compared to the healthy controls (p=0.014), while the DRD1 protein levels were not different between the groups. In control group the genetic variant AA rs267410 (C/A) was associated with a trend of DRD1 protein level reduction in PBL, compared to the CA and CC carriers at the border of statistical significance (p=0.059). In control group minor allele G in homo- and heterozygous state (GG+GC) of rs35916350 (C/G) was associated with a trend to DRD1 mRNA level decrease (p=0.053). Reduction of DRD1 protein level in PBL was detected at haloperidol therapy within 28 days (p=0.001).
