3-нитропропионовая кислота – митохондриальный токсин, ингибирующий сукцинатдегидрогеназу (II комплекс дыхательной цепи переноса электронов). Данный токсин используется для моделирования нейродегенеративных заболеваний. Однако многие процессы, приводящие к повреждению нервной ткани под воздействием 3-нпк, еще не до конца ясны. В связи с этим, целью настоящего исследования было: в экспериментах в системе in vitro и in vivo проверить способность 3-нпк модулировать активность кальций-зависимых цистеиновых протеаз кальпаинов, гиперактивация которых является одним из звеньев нейродегенеративного процесса. Впервые было показано, что аппликация 3-нпк к синаптосомам вызывает активацию m-кальпаина, но не способствует высвобождению кальпаинов из синаптосом; курсовое интраперитонеальное введение 3-нпк крысам приводит к развитию двигательных нарушений, снижению содержания дофамина в стриатуме и увеличению активности кальпаинов (m- и мю) в клетках гиппокампа.
3-nitropropionic acid is a mitochondrial toxin that inhibits succinate dehydrogenase (II complex of the respiratory electron transfer chain). This toxin is used to model neurodegenerative diseases. However, many of the processes that lead to damage neurons under the influence of 3-nitropropionic acid are not yet fully clear. In this regard, the purpose of this study was to test the ability of 3- nitropropionic acid to modulate the activity of calcium-dependent cysteine proteases of calpains in experiments in vitro and in vivo. Their activation is one of the parts of the neurodegenerative process. For the first time, it was shown that the application of 3-nitropropionic acid to synaptosomes causes activation of m-calpain, but does not contribute to the release of calpain from synaptosomes; course intraperitoneal administration of 3-NPC to rats leads to the development of motor disorders, a decrease in the content of dopamine in the striatum and an increase in the activity of calpain (m - and mu) in hippocampal cells.
