Данная работа посвящена созданию биоинженерной конструкции (БИК) и оценке ее регенеративного потенциала на модели патологии эпителия роговицы ex vivo. В результате была получена линия ЛСК. Получен лентивирусный вектор LV-CMV-EGFP Hygro (656–4) и линия ЛСК-EGFP. Создана БИК на основе ЛСК-EGFP. Получена тканевая культура свиных роговиц и разработана модель патологии. Подобраны условия трансплантации ЛСК-EGFP в составе БИК. Получены данные о возможности миграции ЛСК-EGFP на поверхность роговицы. Данная работа являлась неотъемлемым этапом дальнейших исследований регенеративного потенциала ЛСК и создания новых БИК.
This work is devoted to the creation of a bioengineered structure (BIC) and the evaluation of its regenerative potential on an ex vivo model of corneal epithelial pathology. As a result, the LSСline was obtained. The lentiviral vector LV-CMV-EGFP Hygro (656-4) and the LSC-EGFP line were also obtained. The BIC was created based on LSK-EGFP. A pig corneas tissue culture was obtained, and a model of pathology was developed. The conditions of LSC-EGFP transplantation in the BIC were selected. Data on the possibility of LSC-EGFP migration to the corneal surface were obtained. This work was an integral stage of further research of the regenerative potential of LSCs and the creation of new BIC.
