В работе проводятся кристаллографические исследования двух рекомбинантных белков: цитоплазматического домена сигма-1 рецептора и N-концевого SH3 домена белка Grb2. Кристаллизация осуществляется методом «висячей капли». С помощью хроматографии высокого разрешения показа-но, что рекомбинантный цитоплазматический домен сигма-1 рецептора су-ществует в растворе в виде смеси мономерной и нескольких олигомерных форм; кристаллизационные исследования выявили новые условия для его кристаллизации. Получена структура N-концевого SH3 домена белка Grb2 с более высоким разрешением (2,5 Å в сравнении с 3,1 Å). Определена кристаллическая структура n-Src петли, отсутствующая в предыдущих структурах. Получены новые данные о взаимодействии N-концевого SH3 домена с лигандами.
This thesis provides the information about crystallographic research of two recombinant proteins: cytoplasmic domain of sigma-1 receptor and N-terminal SH3 domain of Grb2. The crystallization was performed using the hanging drop technique. By means of high-resolution liquid chromatography it was shown that in saluted state recombinant CTD of sigma-1 receptor exists as a mixture of mono- and oligomers. The crystallographic research shown some new conditions for crystallization of sigma-1 receptor. The structure of N-terminal SH3 domain of Grb2 was successfully solved at higher resolution of 2.5 Å (compared to pre-viously reported 3.1 Å structure). The crystal structure of n-Src loop, which was absent in previously deposited structure, was resolved. Also, some new data about ligand interaction of N-terminal SH3 domain of Grb2 was found.
