Задачи, которые решались в ходе исследования:1) Провести литературный обзор по теме.2) Провести посадку, выращивание и сбор растительного материала для молекулярно-генетических исследований.3) Освоить методику постановки ПЦР в реальном времени (RT PCR).4) Освоить методику анализа данных, полученных в результате проведения RT PCR, с помощью 2-∆CT метода.5) Сделать выводы по полученным результатам.Объекты исследования – растительный и грибной материал:1) Medicago lupulina. В эксперименте была использована линия MlS-1 из сортопопуляции ВИК32 люцерны хмелевидной (Medicago lupulina) [7].2) Plectranthus australis. АМ грибы являются облигатными симбионтами, поэтому для развития АМ гриба (штамм RCAM00320) требуется наличие растения-хозяина. Штамм поддерживается на накопительной культуре плектрантуса южного (Plectranthus australis).3) В качестве АМ-гриба для инокуляции использовался эффективный штамм RCAM00320 Rhizophagus irregularis.В ходе исследований проведена оценка экспрессии генов аквапоринов на 7 различных сроках развития у двух вариантов одной линии MlS-1 из сортопопуляции ВИК32 M. lupulina. Определено, что экспрессия генов некоторых аквапоринов M. lupulina под влиянием АМ-грибов на разных сроках претерпевает существенные изменения. Однако точно выявить паттерны экспрессии на данном этапе исследований достаточно сложно и требуются более детальные исследования по каждому аквапорину в отдельности. В результате работы выявлены гены аквапоринов, изменения экспрессии которых выражены наиболее сильно: NIP2.1, NIP4.2, PIP2.1, SIP1.3, TIP1.1 и TIP4.1. Для каждого из них выдвинуты предположения по влиянию микоризации на экспрессию и в связи с их возможными функциями, описанными в других работах для генов-ортологов на Medicago truncatula.
The tasks of the study:1) Conduct a literary review on the topic;2) Plan, grow and collect plant material for molecular genetic research;3) Master the technique of real-time PCR (RT PCR);4) Master the methodology of analyzing obtained data which is a result of RT PCR using the 2 -∆CT method;5) Draw conclusions based on the results obtained.Objects of research – plant and fungi material:1) Medicago lupulina. In the experiment, the MlS-1 line was used from the varietal population of VIC32 alfalfa hop (Medicago lupulina) [7].2) Plectranthus australis. AM fungi are obligate symbionts, therefore, the presence of a host plant is required for the development of an AM fungus (strain RCAM00320). The strain is maintained on the accumulative culture of the southern plectranthus (Plectranthus australis).3) An effective strain RCAM00320 Rhizophagus irregularis was used as an AM fungus for inoculation.During the research, the expression of aquaporin genes was evaluated at 7 different developmental periods in two different M. lupulina lines. It was determined that the expression of genes of M. lupulina aquaporins under the influence of AM fungi undergoes significant changes at different periods. However, it was quite difficult to accurately identify expression patterns at this stage of research and more detailed studies on each aquaporin separately were required. As a result of the work, the aquaporin genes whose expression was changing were identified: NIP2.1, NIP4.2, PIP2.1, SIP1.3, TIP1.1 and TIP4.1. For each of them, assumptions have been made on the effect of mycorrhization on expression and in connection with their possible functions described in other works for orthologous genes on Medicago truncatula.
