Настоящая магистерская диссертация посвящена изучению роли лектина C-типа DC-SIGN в развитии гриппозной инфекции. В работе приведен обзор литературных данных о строении, организации генома, жизненном цикле вируса гриппа, приведена общая характеристика лектинов С-типа, рассмотрена роль лектина DC-SIGN в гриппозной инфекции. В ходе исследования был разработан и успешно внедрен в лабораторную практику протокол секвенирования полной нуклеотидной последовательности гена DC-SIGN человека, разработана система праймеров для анализа экспрессии мРНК различных изоформ DC-SIGN методом ПЦР в реальном времени с использованием интеркалирующего красителя SYBR Green. Разработанная система позволяет различать мРНК мембранных и растворимых изоформ DC-SIGN, а также мРНК изоформ типов А и B. При экспериментальном заражении дендритных клеток вирусами гриппа А выявлено снижение уровня экспрессии мРНК трансмембранных изоформ DC-SIGN в 50 раз.
This master thesis is devoted to the study the role of C-type lectin DC-SIGN in the progress of influenza virus infection. This work includes review of the literature on structure, organization of influenza genome, lifecycle of the influenza virus, general description of the C-type lectins and possible role of DC-SIGN in the course of influenza virus infection is discussed. Protocol of sequencing of the full-length nucleotide sequence of human DC-SIGN gene was developed and successfully introduced into laboratory practice. Also the real-time PCR assay for the analysis of mRNA expression of various DC-SIGN isoforms was developed. This assay can distinguish mRNAs encoding soluble and membrane isoforms of DC-SIGN and mRNAs of types A and B isoforms. Analysis of DC-SIGN mRNA expression by developed RT-PCR assay showed 50-fold decrease in mRNA level of membrane isoforms in dendritic cells infected by influenza A viruses.
