Целью данной работы являлось изучение влияния красного пигмента, обладающего противоамилоидным действием, на проявление человеческого альфа-синуклеина на модели двух штаммов дрожжей. Эндогенный пигмент, вырабатываемый в данных штаммах благодаря мутации ade1, подавляет проявление гибридного белка альфа-синуклеин-GFP. Это выражается в понижении средней величины флуоресценции GFP, а также доли клеток, накапливающих альфа-синуклеиновые цитоплазматические включения. Экзогенный пигмент отличается от эндогенного по воздействию на альфа-синуклеин. Он вызывает увеличение количества клеток с цитоплазматическими включениями, и практически не влияет на среднюю флуоресценцию. Но, в обоих случаях как эндогенный, так и экзогенный пигменты снижают токсичность альфа-синуклеина в клетках, а также количество белка в осадочной фракции клеточных лизатов.
The aim of this work was to study the effect of the red pigment which exhibit antiamyloid properties, on the model of two yeast strains with different forms of human alfa-synuclein. Endogenous pigment produced in these strains by strains carrying a ade1 mutation inhibits expression of the hybrid protein alfa-synuclein-GFP. This is witnessed by decrease of mean value of GFP fluorescence and by diminishing the per cent of cells exhibiting cytoplasmic inclusions of alfa-synuclein-GFP. Exogenous red pigment differs from the endogenous pigment by it effect on alfa-synuclein. It leads to increase the number of cytoplasmic inclusions and does not change the number of cells expressing GFP fluorescence. Both endogenous and exogenous red pigments lead to drop of toxicity of the cloned alfa-synuclein.
