Пентамидин и диминазин – это два диамидиновых соединения, используемых в качестве противомикробных препаратов. В данной работе показывается, что данные соединения ингибируют NMDA рецепторы концентрационно-зависимым образом. Ингибирование пентамидином является определённо потенциал-зависимым, у диминазина наблюдаются потенциал-зависимая и потенциал-независимая компоненты. Диминазин демонстрирует хвостовые токи и овершуты, что говорит о действии по механизму «foot-in-the-door». Пентамидин же проявляет частичную ловушку в закрытом канале NMDA рецептора. Такое различие в механизме действия может быть объяснено отличиями 3D структуры данных соединений. В молекуле пентамидина две бензамидиновые группы соединены гибким линкером, что позволяет молекуле складываться и помещаться внутри закрытого NMDA канала. Диминазин же имеет жёсткую структуру и не может оказаться в ловушке.
Pentamidine and diminazene are anti-protozoal diarylamidine compounds. In this study we show that they inhibit N‐methyl‐D‐aspartate (NMDA) receptors in a concentration-dependent manner. The inhibition by pentamidine was strongly voltage-dependent, while that of diminazene had voltage-independent components. Diminazene demonstrated tail currents and overshoots suggesting “foot-in-the-door” mechanism of action. In contrast, pentamidine was partially trapped in the closed NMDA receptor channels. Such difference in the mechanism of action can be explained by the difference in the 3D structure of compounds. In the pentamidine molecule two benzamidine groups are connected with a flexible linker, which allows the molecule to fold up and fit in the cavity of a closed NMDA receptor channel. Diminazene, in contrast, has an extended form and could not be trapped.
