Целью настоящей работы являлось исследование структурных различий амилоидных фибрилл на основе лизоцима, полученных при различных условиях фибриллогенеза, и амилоидных фибрилл на основе бета-2-микроглобулина и его укороченных форм без участков полипептидной цепи из 6 и 10 N-концевых аминокислот. В работе использованы методы электронной микроскопии, КД-спектроскопии, а также специально разработанный подход, основанный на абсорбционной и флуоресцентной спектроскопии растворов, амилоидных фибрилл со специфическим флуоресцентным зондом тиофлавином Т, подготовленных методом равновесного микродиализа. Показано что исследуемые амилоидные фибриллы различаются своими фотофизическими характеристиками, морфологией, параметрами связывания с тиофлавином Т и характеристиками связанного красителя, что свидетельствует о полиморфизме этих амилоидных фибрилл.
The aim of this work was to study the structural differences between amyloid fibrils formed from lysozyme obtained under different conditions of fibrillogenesis and amyloid fibrils formed from the full-length β-2-microglobulin as well as its variants with the lack of the protein polypeptide chain of 6 and 10 N-terminal amino acids. The methods of electron microscopy, CD spectroscopy together with a specially developed approach based on the absorption and fluorescence spectroscopy of solutions of amyloid fibrils with a specific fluorescent probe thioflavin T, prepared by the method of equilibrium microdialysis, were used. It was shown that the investigated amyloid fibrils differ in their photophysical properties, morphology, parameters of thioflavin T binding and characteristic of the fibril-bound dye.
