Целью данной работы являлась идентификация и характеристика нового тиазол-оксазол-модифицированного пептида из бактерии Klebsiella pneumoniae subsp. ozaenae, обнаруженного в результате биоинформатического поиска неохарактеризованных генетических кластеров, гомологичных кластеру микроцина В17. Было показано, что при гетерологической экспрессии оперона из K. pneumoniae subsp. ozaenae в клетках E. coli производится вещество, структура которого была частично охарактеризована методами масс-спектрометрии, и показано, что вещество содержит четыре гетероцикла: один оксазольный и три тиазольных, - а также включает дополнительную модификацию на N-конце пептида. Было продемонстрировано, что производимое соединение обладает антибактериальной активностью в отношении E. coli, и активность зависит от присутствия модификации на N-конце пептида и немодифицированных С-концевых аминокислот.
The aim of this work was to identify and describe the structure of a new thiazole-oxazole modified peptide from Klebsiella pneumoniae subsp. ozaenae. Cluster of genes of a new TOMM was found as a result of bioinformatic search of microcin B17 homologs. Detected cluster was introduced into a heterologous expression host, E. coli, which was shown to produce a substance belonging to the class of TOMM. The structure of produced peptide was partially characterized: mass spectrometric analysis revealed that it contains four heterocycles (one oxazole and three thiazole) and also includes an additional modification at the N-terminus of the peptide. We demonstrated that identified cluster of genes is capable of producing a biologically active molecule. Moreover, produced compound was shown to have an antibacterial activity against E. coli. As it was determined this activity depends on the presence of modification at the N-terminus of the peptide and unmodified C-terminal amino acids.
