Предполагается, что нейродегенерация при болезни Паркинсона ассоциирована с гибелью дофаминергических нейронов, обусловленной накоплением олигомерных форм альфа-синуклеина в них. Показано, что на процесс олигомеризации альфа-синуклеина в клетке может влиять дофамин. В то же время олигомерный альфа-синуклеин способствует потере ядерной локализации ключевого фермента в процессе метилирования геномной ДНК, ДНК-метилтрансферазы 1 (DNMT1), приводя к снижению степени метилирования регуляторной области гена SNCA. В связи с этим целью настоящей работы было исследование уровня ДНК-метилтрансферазы 1 в CD45+ клетках периферической крови, а также влияниеinvitroэкзогенного ДА на уровень DNMT1 в лимфоцитах периферической крови при БП. Нами впервые было показано снижение уровня белка DNMT1 в ЛПК пациентов с БП по сравнению с контролем. Показано отсутствие влияния дофамина на уровень DNMT1 в ЛПК пациентов с БП и индивидуумов контроля в ядерной и цитозольной белковых фракциях. Таким образом, полученные данные позволяют предложить использование уровня DNMT1 в ЛПК в качестве биомаркера развития БП.
It is assumed that neurodegeneration in Parkinson's disease is associated with the flexibility of dopaminergic neurons, due to the accumulation of oligomeric forms of alpha synuclein in them. It has been shown that dopamine can influence the process of oligomerization of alpha-synuclein in a cell. At the same time, oligomeric alpha synuclein contributes to the localization of a key enzyme in the process of methylation of genomic DNA, DNA methyltransferase 1 (DNMT1), leading to a low degree of methylation of the regulatory region of the SNCA gene. The purpose of this work was to study the level of DNA methyltransferase 1 in CD45 + cells and the affect of exogenous DA at the level of DNMT1 in peripheral blood lymphocytes in PD. We first showed a decrease in the level of protein DNMT1 in comparison with the control. The absence of dopamine influence on the level of DNMT1 in the PBL of patients with PD and control group individuals in the nuclear and cytosolic protein fractions wasshown.Thus, the data allow us to suggest the use of the DNMT1 level in the diagnostics as a biomarker for the development of PD.
