Болезнь Хантингтона (БХ) и некоторые виды аутосомно-доминантных спиноцеребеллярных атаксий (СЦА) являются самыми известными среди нейродегенеративных полиглутаминовых заболеваний. Как при БХ, так и при СЦА2 наблюдается поражение клеток Пуркинье (КП) коры мозжечка и нарушения моторных функций. Известно, что кальций-активируемые калиевые каналы малой проводимости (SK каналы) осуществляют контроль пейсмейкерной активности КП. Используя метод внеклеточной регистрации, мы проанализировали электрофизиологические функции КП анестезированных мышей-моделей БХ линии YAC128 и бодрствующих мышей-моделей СЦА2 линии SCA2-58Q. В результате систематического введения положительного модулятора SK каналов хлорзоксазона наблюдалось восстановление электрофизиологических функций КП и улучшение синаптической активации КП параллельными волокнами в случае мышей БХ и восстановление нормальной регулярности генерации импульсных сигналов КП в случае мышей СЦА2 и БХ. Мы предполагаем, что SK каналы могут быть рассмотрены в качестве возможной мишени для лечения БХ и СЦА2.
Huntington’s disease (HD) and several types of spinocerebellar ataxias (SCA) are the most common diseases among the neurodegenerative polyglutamine disorders. Dysfunction of cerebellar Purkinje cells (PCs) and impairment of motor functions are observed both in HD and SCA2. It is known that small conductance calcium-activated potassium channels (SK channels) are responsible for the cerebellar PC pacemaker activity. Using an extracellular single-unit recording method we evaluated electrophysiological functions of PCs of anesthetized transgenic mice-models of HD line YAC128 and awake transgenic mice-models of SCA2 line SCA2-58Q of different ages. As a result of systematic administration of positive modulator of SK channels chlorzoxazone, the restoration of electrophysiological functions of PCs and improvement of PCs synaptic activation by parallel fibers in the case of HD mice and restoration of normal firing pattern of PCs in the case of SCA2 and HD mice were observed. We propose that SK channels can be a possible target to treat HD and SCA2.
