Данная работа посвящена изучению потенциал-независимых актин-управляемых натриевых каналов в клетках лейкемии человека К562. По своим характеристикам и механизмам регуляции исследуемые каналы близки к семейству ENaC, за исключением чувствительности к ингибитору амилориду. Остается открытым вопрос об их возможном субъединичном составе. Целью работы являлось изучение возможного участия δ-субъединицы ENaC в составе функционально-активного натриевого канала в клетках К562. В конфигурации whole-cell метода патч-кламп зарегистрировали активность одиночных каналов, активируемых при разборке цитоскелета Цитохалазином Д (100% опытов), изучили их характеристики в Na+ или Li+ содержащем внеклеточном растворе. Проводимость части регистрируемых каналов для Li+ была снижена по сравнению с Na+ (145 мМ). Эта особенность является основным биофизическим признаком ENaC, содержащих δ субъединицу (вместо α). Результаты иммунофлуоресцентного окрашивания подтвердили экспрессию δ-hENaC (наряду с α-, β-, γ-ENaC). В опытах whole-cell обнаружено, что внеклеточное добавление капсазепина, активатора δ-ENaC, вызывало активацию натриевых каналов в 67% опытов. Биофизические характеристики каналов, активированных капсазепином, совпадали с характеристиками актин-управляемых каналов. При замене Na+ в камере на Li+ наблюдали уменьшение амплитуды токов и проводимости каналов. На основании полученных результатов можно предположить, что функционально-активные каналы ENaC в клетках К562 содержат δ субъединицу. Дальнейшее изучение состава и механизмов регуляции ENaC в трансформированных клетках крови актуально для разработки новых таргетных терапевтических стратегий воздействия на раковые клетки.
The given work is devoted to voltage-independent sodium channels in human leukemia cells. According to their characteristics and regulatory mechanisms they are close to the ENaC family, but not sensitive to inhibitor amiloride. The question remains about their possible subunit composition. The aim of the work was to study the possible participation of the δ-ENaC subunit in the composition of functionally active sodium channels in K562 cells. Using whole-cell patch-clamp method and Cytochalasine D for cytoskeleton disassembly, we recorded the activity of single actin-gated channels (in 100% of experiments), their characteristics were studied in extracellular solution containing Na+ or Li+. The single-channel conductance of some channels for Li+ was reduced in comparison to Na+ (145 mM). This characteristic is the main biophysical feature of ENaC that contains δ-subunit (instead of α). Using immunofluorescence staining of K562 cells with specific antibodies, the presence of the δ-subunit of ENaC was confirmed (along with α-, β-, γ-ENaC). In whole-cell experiments it was found that the extracellular addition of capsazepine, an activator of δ ENaC, caused the activation of sodium channels in 67% experiments. The electrophysiological characteristics of the capsazepine- activated channels coincided with the characteristics of actin-gated channels. The replacement of solution in the chamber with Li+-containing one resulted in decrease of current amplitude and channel conductance. Based on this fact, it can be assumed that ENaC channels in K562 cells contain δ subunit. Further study of the composition and mechanisms of regulation of ENaC in transformed blood cells is relevant for the development of new targeted therapeutic strategies for cancer treatment.
