Гигантский бактериофаг phiKZ во время инфекции образует внутри бактерии-хозяина сферическую структуру, состоящую из белковой оболочки и ДНК. Эта структура удерживается в центре бактериальной клетки с помощью тубулино-подобных фаговых белков. Внутри данной структуры были обнаружены белки, участвующие в репликации и транскрипции ДНК бактериофага. Таким образом, данная структура по своей форме и функциям похожа на ядро эукариотической клетки и была названа «псевдо-ядро». Существует гипотеза о том, что вирусы, образующие подобные структуры, могли сыграть важную роль в становлении эукариот. Изучение аспектов функционирования псевдо-ядра является актуальной задачей с точки зрения эволюции. Встаёт вопрос о специфичности упаковки ДНК внутрь данной структуры. Для ответа на этот вопрос в первую очередь необходимо определить, происходит ли деградация ДНК бактерии во время инфекции. В данной работе с помощью методов электрофоретического разделения ДНК и ПЦР в реальном времени было установлено, что ДНК бактерии не подвергается деградации при инфекции бактериофагом phiKZ.
Giant bacteriophage phiKZ forms a spherical structure, containing a protein shell and DNA, in its host bacteria cell during infection. This structure is positioned in the center of the cell by tubulin-like phage proteins. Some protein participating in replication and transcription of phage DNA were found inside this structure. Therefore, this structure by its organization and function resembles a nucleus of a eukaryotic cell and was called “pseudo-nucleus”. A hypothesis was proposed that viruses that create similar structures in ancient cells may have played an important role in evolution of eukaryotes. Studying the functioning of pseudo-nucleus is an important problem for an evolutionary point of view. First arises a question about specificity of DNA packing into the pseudo-nucleus. To answer that question, it needs to be determined whether bacterial DNA undergoes degradation in an infected cell. In this work DNA gel-electrophoresis and real-time PCR were used to establish that bacterial DNA does not undergo degradation during the infection of phiKZ bacteriophage.
