Данная работа посвящена исследованию возможного механизма ингибирования деления клеток Escherichia coli, находящихся в состоянии SOS-ответа и измерению минимальной концентрации белка SulA, достаточной для эффективного ингибирования клеточного деления. Для этого в ходе работы была создана и использована конструкция, позволяющая имитировать SOS-ответ в клетках E. coli в виде остановки деления. Для измерения концентрации белка использовали независимые методы, такие как анализ концентрации по измерению интенсивности флуоресценции и иммуноблоттинг. В результате были получены средние значения концентрации белка SulA, достаточной для ингибирования деления. Сравнение концентрации SulA в клетке с известной концентрацией белка FtsZ позволило установить, что для ингибирования достаточно субстехиометрических количеств SulA. Полученные результаты говорят о том, что существующая на сегодняшний день модель секвестрации не полностью объясняет механизм ингибирования деления белком SulA in vivo в условиях данного эксперимента. Будущие исследования должны помочь установить, какая модель блокирования деления является наиболее адекватной, и, следовательно, лучше понять механизмы SOS-ответа, который позволяет бактериям пережить неблагоприятные для них условия.
The given work is devoted to study the mechanism of cell division inhibition in Escherichia coli during SOS response and to measure the minimal concentration of SulA protein in the cell, which is sufficient to block the cell division. During this work a construction was created and used which allowed us to mimick the SOS response in E. coli cells in terms of the cell division arrest. Measurements of protein concentration were carried out using several independent methods, such as fluorescent intensity quantification or semi-quantifying Western blotting. As a result, SulA concentration values sufficient to inhibit division were determined. Comparison between obtained concentration of SulA and concentration of FtsZ well-known from publications showed that it is enough to add substoichiometric quantity of SulA for blocking cell division. The results indicate that the sequestration model is not comprehensive to explain in vivo mechanism of blocking division by the action of the SulA protein under the conditions of this experiment. Further study should help to find out which model is correct, and, thereafter, to better explain the action of SOS response mechanism, which allows bacteria to survive the adverse conditions.
