Тема выпускной квалификационной работы: «Редокс-метаболизм мезенхимных стволовых клеток человека при развитии стресс-индуцированного старения». В данной работе проанализировано изменение концентрации внутриклеточной перекиси водорода в мезенхимных стволовых клетках эндометрия (МСКЭ) человека при индукции клеточного старения. Методическая основа работы – использование генетически кодируемого биосенсора перекиси водорода HyPer, а также рН-биосенсора SypHer. Задачи, которые решались в ходе исследования: 1. Трансдуцировать МСКЭ лентивирусами для генерации клеточных линий, стабильно-экспрессирующих биосенсоры HyPer и SypHer в клеточной цитоплазме. 2. Сравнить характеристики клеток, экспрессирующих биосенсор HyPer, при стресс-индуцированном и репликативном старении. 3. Провести оценку редокс-состояния постаревших клеток. Работа проведена на базе ФГБУ "Институт цитологии РАН". В работе показано, что все основные признаки старения, индуцируемые окислительным стрессом в культурах МСКЭ, экспрессирующих HyPer, соответствуют характеристикам репликативно состаренных клеток. Обнаружено, что при развитии стресс-индуцированного старении клеток наблюдается увеличение уровня активных форм кислорода и закисление клеточной цитоплазмы, однако, увеличения концентрации перекиси водорода в цитоплазме клеток не происходит. Полученные данные могут иметь фундаментальное значение для развития теорий клеточного старения.
The theme of the final qualification work: "Redox metabolism of human mesenchymal stem cells during the development of stress-induced aging." In this paper, we analyzed the change in the concentration of intracellular hydrogen peroxide in human endometrial mesenchymal stem cells (MSCE) during the induction of cellular aging. The methodological basis of the work is the use of the genetically encoded biosensor of hydrogen peroxide HyPer, as well as the pH biosensor SypHer. Tasks that were solved during the study: 1. Transduce MSCE with lentiviruses to generate cell lines stably expressing HyPer and SypHer biosensors in the cell cytoplasm. 2. Compare the characteristics of cells expressing biosensors in stress-induced and replicative aging. 3. Assess the redox state of the aged cells. The work was carried out based on the Institute of Cytology RAS. It was shown that all the main signs of aging-induced by oxidative stress in MSCE cultures expressing HyPer correspond to the characteristics of replicative aged cells. It was found that with the development of stress-induced cell aging, an increase in the level of reactive oxygen species and acidification of the cell cytoplasm is observed, however, an increase in the concentration of hydrogen peroxide in the cell cytoplasm does not occur. The findings may be fundamental to the development of theories of cell aging.
