В данной работе проанализирована возможность использования наноразмерных частиц комплексного состава для внутриклеточной доставки нуклеиновых кислот. Исследуемые носители разрабатываются с целью последующего применения в терапии возрастных и наследственных дегенеративных изменений сетчатки. В ходе выполнения работы была исследована эффективность ассоциации/интернализации полученных частиц на модельных клеточных линиях in vitro и на модели здоровых животных in vivo. Эффективность ассоциации для клеточной линии SH-SY5Y, моделирующей сетчатку глаза, составила до 88% при незначительном токсическом воздействии. В эксперименте in vivo, путем анализа гистологических препаратов, было выявлено, что капсулы размером ~200 нм являются эффективными переносчиками генетического материала (мРНК, кодирующая зеленый флуоресцентный белок).
In this work, the possibility of using polymer calcium carbonate particles as a delivery system for nucleic acids in gene therapy of retinal degeneration was analyzed. The studied carriers are being developed for treatment of age-related and hereditary retinal degenerative changes. The effectiveness of association/internalization of these carriers on model cell lines in vitro and on healthy animal models in vivo was researched. The efficiency of the cell / particle association for the SH-SY5Y cell line modeling the retina is 88% with minimal toxic effects. It was revealed that capsules with a size of ~200 nm are effective carriers of genetic material (mRNA encoding a green fluorescent protein) by analyzing histological preparation.
