Данная работа посвящена изучению механизма индуцированной хондрогенной дифференцировки мезенхимных стромальных клеток человека, полученных из десквамированного эндометрия в менструальной крови здоровых доноров (эМСК). Цель исследования – продемонстрировать возможность хондрогенной дифференцировки в 2D-культуре эМСК и изучить зависимость ее эффективности от активности MAP- и PI3-киназных сигнальных каскадов. Среди экспериментальных методов, использованных в работе: культивирование клеток, экстракция мРНК из клеточных лизатов, электрофорезы (белковый, РНК и ДНК), иммуноблотинг, ОТ-ПЦР в режиме реального времени, а также окрашивание клеточных препаратов алциановым синим и сафранином О. В результате проведенной работы была показана возможность хондрогенной дифференцировки эМСК в монослое (2D-культуре) и динамика этого процесса в течение 14-ти дней; также была выявлена зависимость эффективности дифференцировки от активности MAP- и PI3-киназных сигнальных каскадов, что дает представление о молекулярном механизме процесса. Вышеописанные феномены составляют научную новизну представленного исследования.
This work is devoted to the study of the mechanism of induced chondrogenic differentiation of human mesenchymal stromal cells obtained from desquamated endometrium in the menstrual blood of healthy donors (eMSCs). The aim of the study was to demonstrate the possibility of chondrogenic differentiation in 2D culture of eMSCs and to study the dependence of its effec-tiveness on the activity of MAP-and PI3-kinase signaling cascades. The experimental methods used in this work include cell culture, mRNA extraction from cell lysates, electrophoresis (protein, RNA, and DNA), immunoblotting, real-time RT-PCR, and staining of cell preparations with alcyan blue and safranin O. As a result of this work, the possibility of chondrogenic differentiation of eMSCs in a monolayer (2D culture) and the dynamics of this process over 14 days were shown; the dependence of the differentiation efficiency on the activity of MAP - and PI3-kinase signaling cascades was also revealed, which gives an idea of the molecular mechanism of the process. The above-described phenomena constitute the scientific novelty of the presented research.
