В данной работе оценивалось влияние ГАМКВ рецепторов на протекание эпилептиформной активности в 4-аминопиридиновой модели in vitro. Для достижения поставленной цели был проведен ряд экспериментов: регистрация эпилептиформной активности, анализ парного отношения вызванных возбуждающих постсинаптических токов (ВПСТ), и миниатюрных событий. Было обнаружено, что активация ГАМКB рецепторов (ГАМКВ-Р) оказывает выраженное противоэпилептическое действие в 4-аминопиридиновой модели in vitro, снижая длительность и частоту иктальных разрядов. Агонист ГАМКВ-Р SKF97541 вызывал уменьшение амплитуды постсинаптических ответов и увеличение значения коэффициента парных импульсов, что свидетельствует о его значительном влиянии на пресинаптически-локализованные рецепторы. При этом тест с парными импульсами не показал достоверных изменений в амплитуде ответов и парном отношении при аппликации блокатора ГАМКВ-Р CGP55845, что говорит о низкой фоновой активности пресинаптических ГАМКВ рецепторов. При записи миниатюрных событий ни агонист, ни антагонист ГАМКВ-Р не изменяли амплитуду, время нарастания и τ спада миниатюрных возбуждающих постсинаптических токов (мВПСТ). Однако, CGP55845 увеличивал частоту возникновения мВПСТ, а SKF97541 уменьшал её.
In this work, the influence of GABAB receptors on the epileptiform activity in a 4-aminopyridine model of epilepsy in vitro was evaluated. To achieve this goal, several experiments were carried out: the registration of the epileptiform activity, paired pulse test, and the miniature events analysis. It was found that the activation of GABAB-receptors (GABAB-R) has a pronounced antiepileptic effect in the 4-aminopyridine model in vitro, reducing the duration and frequency of the ictal discharges. The GABAB-R agonist SKF97541 reduced the amplitude of the postsynaptic responses and increased the paired pulse ratio (PPR), which indicates its significant effect on the presynaptic GABAB receptors. In contrast, the application of the GABAB-R blocker CGP55845 did not affect the paired pulse ratio of the postsynaptic current amplitudes, which suggest the low levels of background activity of the presynaptic GABAB receptors. Miniature event analysis have shown that neither the GABAB-R agonist, nor the antagonist, changed the amplitude, rise time, or decay τ of the miniature excitatory postsynaptic currents (mEPSC). However, CGP55845 increased the mEPSC frequency, while SKF97541 decreased it.
