Данная работа посвящена исследованию влияния экзогенного гемина и гематина на эритроциты человека. Гемин (координационный ион – Cl) и гематин (координационный ион – OH) являются окисленными производными гема, которые образуются в организме человека при малярии, геморрагическом инсульте, язве желудка, серповидно-клеточной анемии и бета-талассемии. Эритроциты являются одним из главных объектов действия гемина и гематина, которые способны повреждать мембрану эритроцитов и изменять конформацию цитоскелетных белков. Методом лазерной дифракции было показано, что гемин и гематин индуцируют доза-зависимую сферизацию эритроцитов и их гемолиз. Наблюдаемые эффекты отличались, вероятно, из-за разного лигандирования данных соединений. Скорости сферизации и гемолиза при действии гематина была выше, чем гемина, что говорит о более высокой реакционной способности данного соединения. Также была показана защитная роль альбумина от токсических производных гема. Альбумин, связываясь с гемином и гематином, ингибировал сферизацию и гемолиз эритроцитов. Таким образом, полученные данные позволят лучше понять механизмы действия гемина и гематина на эритроциты человека, что в дальнейшем способствует улучшению стратегий лечения таких заболеваний как малярия, геморрагический инсульт, язва желудка, серповидно-клеточная анемия и бета-талассемия.
The given work is devoted to investigation of the effects of exogenous hemin and hemin on human erythrocytes. Hemin and hematin are oxidized heme derivatives formed in the human organism in malaria, hemorrhagic stroke, gastric ulcer, sickle cell anemia and beta-thalassemia. Erythrocytes are one of the main objects of hemin and hematin action resulting in damage of the erythrocytes membrane and alterations of the cytoskeletal proteins conformation. Using laser diffraction method, it was shown that hemin and hematin induced dose-dependent erythrocyte spherization and hemolysis. The effects of these compounds differed probably because of different liganding. The rates of spherization and hemolysis under the action of hemin were higher than hemin rates, indicating a higher reactivity of this compound. The protective role of albumin against toxic heme derivatives was also shown. Albumin, binding to hemin and hematin, inhibited spherization and hemolysis of erythrocytes. Thus, the obtained data will lead to a better understanding of the mechanisms of action of hemin and hemin on human red blood cells, which further improves treatment strategies for such diseases as malaria, hemorrhagic stroke, gastric ulcer, sickle cell anemia and beta-thalassemia.
