Данная работа посвящена исследованию действия производных фенитоина на AMPA-рецепторы. Фенитоин является основным противосудорожным препаратом, который обратимо ингибирует AMPA-рецепторы с IC50 250 ± 60 мкМ для CP-AMPAR и IC50 30 ± 4 мкМ для CI-AMPAR. Данные были получены методом патч-кламп в конфигурации «целая клетка». Из ряда производных фенитоина наиболее активными оказались 3,3-diphenyl-2,5-pyrrolidinedione и 5-cyclohexyl-5-phenyl-2,4-imidazolidinedione, ингибирующие CI-AMPA рецепторы с IC50 26 ± 2 мкМ и с IC50 11 ± 1 мкМ, соответственно. На основании полученных данных была сделана оценка значимости структурных элементов молекулы фенитоина в контексте его ингибирующего действия. Были выделены наиболее значимые элементы и было определено, что некоторые из них не оказывают существенного влияния на процесс ингибирования. Это исследование может помочь улучшить эффективность и безопасность применения фенитоина, а также может быть использовано для разработки новых противоэпилептических препаратов, разработанных на основе фенитоина.
The given work is devoted to a study of the action of phenytoin derivatives on AMPA receptors. Phenytoin is a major anticonvulsant drug that reversibly inhibits AMPA receptors with an IC50 of 250 ± 60 μM for CP-AMPAR and an IC50 of 30 ± 4 μM for CI-AMPAR. The results were obtained by the patch-clamp method in a whole-cell configuration. Of the series of phenytoin derivatives, 3,3-diphenyl-2,5-pyrrolidinedione and 5-cyclohexyl-5-phenyl-2,4-imidazolidinedione were the most active, inhibiting CI-AMPA receptors with IC50 26 ± 2 μM and with IC50 11 ± 1 μM, respectively. On the basis of the data obtained, the significance of the structural elements of the phenytoin molecule in the context of its inhibitory action was evaluated. The most important elements were highlighted and it was determined that some of them do not have a significant effect on the inhibitory process. This study can help improve the efficacy and safety of phenytoin and can also be used to develop new antiepileptic drugs based on phenytoin.
