Данная работа посвящена разработке и апробации метода локальной фиксации потенциала (патч-кламп) для исследования нативных ионных каналов в плазматической мембране мезенхимных стволовых клеток эндометрия (эМСК) человека при 3D культивировании. Тематика научного исследования является важной в рамках вопроса выявления функциональной активности ионных каналов и их регуляции в физиологических реакциях трехмерных клеточных культур. Основной экспериментальный подход - метод патч-кламп в конфигурации cell-attached, позволяющий записывать ионные токи одиночных каналов в изолированном участке плазматической мембраны клеток. Для регистрации активности ионных каналов трехмерных клеточных агрегатов нами был разработан новый подход для образования стабильного сверхплотного контакта между регистрирующей пипеткой и фрагментом мембраны клеток, агрегированных в сфероиды. С помощью данного протокола нами впервые была зарегистрирована активность механочувствительных ионных каналов (МЧ) в клетках эМСК, собранных в сфероиды. Более того, мы наблюдали их функциональное взаимодействие с калиевыми каналами большой проводимости (BK). С помощью иммунофлуоресцентного окрашивания была показана экспрессия BK каналов в мембране клеток эМСК, агрегированных в сфероиды. Также мы наблюдали достоверное снижение частоты активации МЧ каналов в сфероидах, что может свидетельствовать о различиях в уровне функциональной экспрессии каналов в 3D культуре по сравнению с 2D культурой эМСК.
The present work is devoted to development and application of the specific variant of the patch-clamp method for the recording of the activity of native ion channels in 3D spheroids formed from human endometrial mesenchymal stem cells (eMSCs). The research area is important in terms of registration of the functional activity of ion channels and their regulation in the physiological reactions in 3D cell cultures. The main experimental approach is the patch-clamp method in the cell-attached configuration (registration of single ion channels). To register the ion channel activity in three-dimensional cell aggregates, we developed a new approach for the formation of the stable contact between recording pipette and plasma membrane fragment of the cells aggregated into spheroids. Using the developed approach, we have recorded the activity of mechanosensitive ion channels eMSCs assembled in spheroids. Moreover, we observed their functional interplay with potassium channels of big conductance (BK) in the plasma membrane. Using immunofluorescent staining the BK expression in the membrane of eMSCs was shown. Also, we observed a significant decrease in the frequency of mechanosensitive channel activation in spheroids that may indicate the differences in the level of functional expression of channels in 3D culture comparing to 2D culture of eMSCs.
