Методы молекулярно–генетического анализа получили широкое распространение в различных областях. Однако, для них требуется пробоподготовка, обычно заключающаяся в лизисе пробы и выделении из нее нуклеиновых кислот. Микрофлюидные устройства позволяют объединить часть или все стадии анализа на одном конструктиве и автоматизировать эти процессы, исключив влияние человеческого фактора на результат. Поэтому целью работы являлась разработка прототипа устройства пробоподготовки жидких сред, предполагающего последующее объединение с устройством для проведения молекулярно–генетического анализа. Было разработано и апробировано два варианта устройства, предназначенных для использования набора реагентов «М–Сорб–ООМ» (ООО «Синтол»). Прототипы устройств созданы из силиконовой резины PlatSet 40 методом «мягкой» литографии с последующей герметизацией кислородной плазмой. Апробация устройств проведена на примере выделения нуклеиновых кислот из клеток E.coli. Продемонстрирована работоспособность второго варианта устройства. Однако, концентрация выделяемых существенных кислот уступает полученной при ручном выделении (пороговые циклы ПЦР–РВ составили цифры и цифры, соответственно). Это, вероятно связано с сорбцией на стенках каналов. Для ее снижения необходим обработать устройство перед использованием, например, бычьим сывороточным альбумином.
Methods of molecular genetic analysis are widely used in various fields. However, they require sample preparation, usually consisting in lysis of the sample and isolation of nucleic acids from it. Microfluidic devices allow you to combine part or all of the analysis stages on one construct and automate these processes, eliminating the influence of the human factor on the result. Therefore, the aim of the work was to develop a prototype of a sample preparation device for liquid media, involving subsequent integration with a device for molecular genetic analysis. Two versions of the device designed for use with the «M–Sorb–OOM» reagent kit (Syntol LLC) were developed and tested. Prototypes of the devices were created from silicone rubber PlatSet 40 by the method of "soft" lithography followed by sealing with oxygen plasma. The devices were tested using the example of the isolation of nucleic acids from E.coli cells. The operability of the second version of the device is demonstrated. However, the concentration of the essential acids released is inferior to that obtained by manual isolation (the threshold cycles of PCR–RV were figures and figures, respectively). This is probably due to sorption on the walls of the channels. To reduce it, it is necessary to treat the device before use, for example, with bovine serum albumin.
