Тема выпускной квалификационной работы: «Определение генетических детерминант и фенотипических характеристик гипервирулентности Klebsiella pneumoniaе» (K. pneumoniae). Данная работа посвящена изучению различных генов вирулентности клинических изолятов K. pneumoniae, а также фенотипических свойств гипервирулентных штаммов.Задачи исследования – изучение фенотипических характеристик вирулентности у выделенных штаммов K. pneumoniae, составление перечня генетических детерминант, связанных с гипервирулентность, исследование генетических детерминант вирулентности у выделенных штаммов клебсиелл и разработка мультиплексной ПЦР для одновременного обнаружения генов, связанных с гипервирулентным фенотипом.Объект исследования – 65 клинических изолятов K. pneumoniae из проб биоматериала от госпитализированных пациентов Санкт-Петербурга. Работа выполнена на базе ФБУН НИИЭМ им. Пастера. Методами изучения изолятов являлись: «Стринг-тест», ПЦР-детекция генов вирулентности и электрофорез.По результатам работы исследовано 65 клинических изолятов K. pneumoniae из проб биоматериала от госпитализированных пациентов Санкт-Петербурга и разработана мультиплексная ПЦР для одновременного обнаружения трех генов вирулентности uni-rmpА, iroB, iutA.
The subject of the graduate qualification work is "Determination of genetic determinants and phenotypic characteristics of Klebsiella pneumoniae hypervirulence". This work is devoted to the study of various virulence genes of K. pneumoniae clinical isolates, as well as the phenotypic properties of hypervirulent strainsThe tasks of the study: To make a research of phenotypic virulence characteristics in isolated K. pneumoniae strains, to make a list of genetic determinants associated with hypervirulence, to study genetic determinants of virulence in isolated Klebsiella strains and to conduct a multiplex PCR for simultaneous detection of genes associated with hypervirulent phenotype. The object of the study is 65 clinical K. pneumoniae isolates from biomaterial samples from hospitalized patients in St. Petersburg.The work was carried out on the basis of the Saint-Petersburg Pasteur Institute The methods of studying isolates were: "String-test", PCR detection of virulence genes and electrophoresis.In the course of the research, 65 clinical K. pneumoniae isolates were studied from biomaterial samples from hospitalized patients in St. Petersburg and multiplex PCR was developed for simultaneous detection of three virulence genes - uni-rmpА, iroB, iutA.
