Гигантские бактериофаги, родственные phiKZ, осуществляют транскрипцию своего генома при помощи двух собственных многосубъединичных ДНК-зависимых РНК-полимераз: вирионной (вРНКП) и невирионной (нвРНКП). НвРНКП отвечает за транскрипцию поздних генов. По-видимому, регуляция транскрипции поздних генов осуществляется по-разному для каждого вида гигантских бактериофагов. Бактериофаг phiKZ на поздних стадиях инфекции формирует внутри инфицированной клетки псевдо-ядро, где осуществляется транскрипция фагового генома, в то время как геном бактериофаг AR9 распределён в цитозоле клетки в течение всего литического цикла. В данной работе мы применили методы биоинформатики для исследования гипотезы о наличии взаимосвязи ДНК-связывающих свойств нвРНКП, кодируемых phiKZ-родственными гигантскими бактериофагами, и особенностей морфологии инфицированной клетки. Был проведён филогенетический анализ субъединиц нвРНКП бактериофагов phiKZ, AR9 и белка псевдо-ядра. Результатом работы стало подтверждение рассматриваемой гипотезы.
Jumbo bacteriophages related to phiKZ transcribe their genome using two of their own multisubunit DNA-dependent RNA polymerases: virion (vRNAP) and non-virion (nvRNAP). NvRNAP is responsible for the transcription of late genes. Apparently, the regulation of transcription of late genes is carried out differently for each species of giant bacteriophages. The bacteriophage phiKZ forms a pseudo-nucleus inside the infected cell at the late stages of infection, where the transcription of the phage genome is carried out, whereas the bacteriophage AR9 genome is distributed in the cytosol of the cell during the entire lytic cycle. In this work, we applied bioinformatics methods to investigate the hypothesis about the presence of a relationship between the DNA-binding properties of nvRNAP encoded by phiKZ-related jumbo bacteriophages and the features of the infected cell morphology. Phylogenetic analysis of nvRNAP subunits of bacteriophages phiKZ, AR9 and pseudo-nucleus protein was carried out. The result of the work was the confirmation of the considered hypothesis.
