Данная работа посвящена изучению двух ранних генов бактериофага phiKZ с целью поиска белков, ингибирующих рост клеток бактерий. В ходе работы была осуществлена сборка экспрессионных плазмид, кодирующих белки, слитые с аффинными полипептидами. Было проведено тестирование влияния экспрессии генов gp103 и gp113 на жизнедеятельность бактерий. Первый этап тестирования заключался в высевании бактерий, содержащих плазмиды с генами, кодирующими исследуемые белки, в присутствии и отсутствии индуктора. Второй этап тестирования заключался в анализе изменений оптических плотностей культур клеток в зависимости от содержания индуктора в среде. В ходе экспериментов был выявлен один белок-кандидат, проявляющий ингибирующую рост активность в отношении бактерий P. aeruginosa и E. coli. Выделение с помощью металло-обменной хроматографии позволило подтвердить наличие белка Gp103 внутри клеток. Благодаря обнаруженным бактерицидным свойствам белка Gp103, данное открытие имеет большой потенциал применения в лечении инфекций, вызванных грамотрицательными P. aeruginosa и E. coli.
The given work is devoted to the study of two early bacteriophage genes phiKZ in order to find proteins that inhibit the growth of bacterial cells. In the course of the work, was carried out the assembly of expression plasmids encoding proteins fused with affine polypeptides. The effect of gp103 and gp113 gene expression on the vital activity of bacteria was tested. The first stage of testing consisted in seeding bacteria containing plasmids with genes encoding the studied proteins in the presence and absence of an inductor. The second stage of testing consisted in the analysis of changes in the optical densities of cell cultures depending on the content of the inductor in the medium. During the experiments, one candidate protein was identified that exhibits growth-inhibiting activity against P. aeruginosa and E. coli bacteria. Isolation by metal-exchange chromatography made it possible to confirm the presence of Gp103 protein inside cells. Due to the discovered bactericidal properties of the Gp103 protein, this discovery has great potential for use in the treatment of infections caused by gram-negative P. aeruginosa and E. coli.
