Растительные экстраклеточные везикулы (РЭВ) являются перспективными препаратами для лечения повреждений кожи. Целью данной исследовательской работы стала оценка влияния РЭВ на регенерацию клеток кожи человека после их механического повреждения in vitro. РЭВ были выделены методом последовательного дифференциального ультрацентрифугирования из микроводорослей, хламидомонады (Chlamydomonas reinhardtii CC125) и хлореллы (Parachlorella kessleri IPPAS c15), а также из плодов томата и огурца. Клетки линий HaCat (кератиноциты), DF-2 (фибробласты), HEK293T (клетки эмбриональной почки человека) в монослое повреждали и со-культивировали с РЭВ в течение 24 часов. Анализ влияния РЭВ на пролиферативную активность неповрежденных клеток (цитотоксичность) проводили с использованием RTCA xCELLigence. Жизнеспособность клеток, подверженных механическому повреждению, оценивали с помощью резазурин-теста. Экспрессию генов, белковые производные которых участвуют в заживлении раны: HAS1, HAS2, ELN оценивали методом ОТ-ПЦР. На основе результатов работы, можно заключить, что РЭВ томата, огурца, хлореллы и хламидомонады не оказывали цитотоксического и онкогенного действия, увеличивали экспрессию таргетных генов в клетках кожи человека после их механического повреждения и повышали их жизнеспособность. В совокупности, РЭВ рассмотренных в данной работе продуцентов потенциально могут оказывать терапевтическое действие при заживлении раны.
Plant extracellular vesicles are promising therapeutics for treating skin injuries. The aim of this research work was to evaluate the effect of PEVs on the regeneration of human skin cells after their mechanical damage in vitro. PEVs were isolated by sequential differential ultracentrifugation from microalgae, chlamydomonas (Chlamydomonas reinhardtii CC125) and chlorella (Parachlorella kessleri IPPAS c15), as well as from tomato and cucumber. HaCaT (keratinocytes), DF-2 (fibroblasts), and HEK293T (human embryonic kidney cells) cell monolayers were scratched and co-cultured with PEVs for 24 hours. The effect of PEVs on the proliferative activity of intact cells (cytotoxicity) was analyzed using RTCA xCELLigence. Cell viability of mechanically damaged cells was assessed using the resazurin assay. The expression of genes whose protein derivatives are involved in wound healing: HAS1, HAS2, ELN was evaluated by RT-PCR. Based on the results of the work, it can be concluded that tomato, cucumber, chlorella and chlamydomonas PEVs did not have a cytotoxic or oncogenic effect, increased cell viability and the expression levels of target genes in human skin cells after their mechanical damage. In conclusion, PEVs considered in this work can potentially have a therapeutic effect in wound healing.
