Данная работа посвящена установлению роли внеклеточного матрикса как составляющей микроокружения опухолевых клеток на их прогрессию. В ходе исследования был отработан протокол получения внеклеточного матрикса раковых клеток остеосаркомы. С помощью ПЦР-тестов в режиме реального времени было установлено повышение экспрессии генов, отвечающих за механизмы сохранения ДНК, аутофагию, эпителиально-мезенхимальный переход, миграцию и пролиферацию клеток, инкубированных на матриксе по сравнению с контрольной линией. Кроме того, полученные результаты метаболических характеристик были подтверждены тестами на пролиферацию клеточных культур. Методом western-blot была оценена активация защитных механизмов культуры клеток HOS после взаимодействия с ВКМ по накоплению белков, участвующих в шаперонной и аутофагической системах. Также был проведен МТТ-тест, свидетельствующий об устойчивости к химиопрепаратам клеток, инкубированных на внеклеточном матриксе, по сравнению с контрольной группой. Таким образом, результатом исследования является понимание взаимодействий внеклеточного матрикса с клетками, влияний на передачу сигналов в клетки. Данная работа может быть полезна для разработки новых стратегий лечения онкологических заболеваний на основе ВКМ.
This work is devoted to establishing the role of the extracellular matrix (ECM) as a component of the microenvironment of tumor cells on their progression. During the study, a protocol for obtaining ones own extracellular matrix from osteosarcoma cancer cells was developed and tested. Real-time PCR tests revealed an increase in the expression of genes responsible for DNA retention mechanisms, autophagy, epithelial-mesenchymal transition, migration and proliferation of cells incubated on the matrix compared to the control line. In addition, the obtained metabolic characterization results were confirmed by cell culture proliferation tests. The western blot method was used to evaluate the activation of protective mechanisms of HOS cell culture after interaction with the ECM by the accumulation of proteins involved in the chaperone and autophagic systems. An MTT test was also performed, indicating the resistance to chemotherapy of cells incubated on the extracellular matrix compared to the control group. Thus, the result of the study is an understanding of the interactions of the extracellular matrix with cells and the effects on signal transmission into cells. This work may be useful for the development of new strategies for treating cancer diseases based on ECM.
