Данная работа посвящена получению и характеристике реассортантного кандидатного вакцинного штамма А/СПб33/22/PR8/1:7/2023 (H1N1pdm09) (RA-56) с последующим получением прототипа ИГВ на его основе. Реассортантный штамм получен методом классической генетики на основе эпидемического штамма А/Санкт-Петербург/НИИГ-33/2022 и универсального высокорепродуктивного донора A/Puerto Rico/8/34/LGV. Показано, что полученный штамм обладает высокой репродуктивной активностью в системе развивающихся куриных эмбрионов и стабильной формулой генома 7:1 с гемагглютинином от штамма дикого типа. Полученный реассортантный штамм обладает фенотипическими свойствами необходимыми для разработки ИГВ, однако отсутствие температурочувствительного фенотипа делает полученный штамм полностью непригодным для разработки ЖГВ. На основе реассортантного штамма изготовлен прототип ИГВ и показана его биологическая безопасность на мышиной модели.
The given work is devoted to development and characterization of candidate reassortant influenza vaccine strain А/СПб33/22/PR8/1:7/2023 (H1N1pdm09) (RA-56) with subsequent development of prototype of inactivated influenza vaccine based on it. Reassortant influenza virus was created using wild-type strain А/Санкт-Петербург/НИИГ-33/2022 and universal high-yield donor strain A/Puerto Rico/8/34/LGV with method of classic genetics. It is shown that reassortant virus is high-yielding in embryonated chicken eggs and possesses stable set 7:1 genome formula with hemagglutinin inherited from wild-type parent strain. It is demonstrated that RA-56 has phenotype suitable for the development of inactivated influenza vaccine, yet lack of temperature-sensitivity makes it impossible to use RA-56 in the development of LAIV. Prototype of inactivated influenza vaccine was created on the basis of RA-56 and its biological safety was evaluated in mice model.
