The given work is devoted vaccine prophylaxis of infections caused by Group A Streptococcus (GAS). The aim of the work was to evaluate the protective efficacy of a vaccine candidate containing immunogenic fragments of C5a-peptidase (ScpA) and the erythrogenic exotoxin SpeA in GAS infection mouse models. The study employed methods of recombinant protein production in the E. coli system, modeling of invasive infection in laboratory animals, as well as molecular genetic approaches (PCR screening of the speA gene, emm typing). As a result, accelerated clearance of GAS M49 in immunized animals was demonstrated using the bacterial clearance model and universal primers for the detection of all speA gene allelic variants were developed. A comprehensive evaluation of the protective efficacy of the chimeric ScpA-SpeA protein was performed for the first time. The obtained data can serve as a basis for further research on the vaccine candidate and optimization of GAS infection models, as well as for epidemiological monitoring of circulating toxigenic GAS strains. The following information technologies were used in this study: MSWord, MS Excel, STATISTICA, NCBI BLAST, BioEdit Sequence Alignment Editor (version 7.2.5), PubMLST, PubMed.
