The given work is devoted to investigations of the effect of medium-chain triglycerides (MCTs) on cognitive impairments in adult male Wistar rats in a model of neonatal inflammation (LPS, 25 μg/kg on days 15, 18, and 21 of postnatal development). Adult animals received 3 g/kg MCFA daily for 28 days. Working and spatial memory, behavior, blood metabolite and cytokine levels, and the expression of Mmp9, Timp1, and NMDA/AMPA receptor subunits in the cortex and hippocampus were assessed. MCFA improved working memory in animals without inflammation, normalized the pyruvate/lactate ratio, modulated IL-10, TNFα, CX3CL1, and IP-10, reduced Mmp9 expression in the hippocampus, and reorganized the composition of glutamate receptors during inflammation. In contrast to similar experiments in female rats, effects of MCFA on behavioral, immune, and transcriptional levels were observed in males, suggesting the existence of sex differences in the response to neonatal inflammation and the consumption of MCFA triglycerides over a course of treatment.
