Целью работы было изучить влияние суб-цитотоксических доз антиоксидантов на протекание пролиферативных фаз клеточного цикла в синхронизованных по клеточному циклу культурах мезенхимных стволовых клетках эндометрия человека. Установлено, что добавление субцитотоксических доз антиоксидантов в ростовую среду МСКэ на стадии ранней фазы синтеза ДНК приводит к дозо-зависимому замедлению синтеза ДНК и блоку клеточного цикла в G2/M-фазе. Анализ накопления маркеров разрывов ДНК - фосфорилированной модификации гистона H2AX и активной АТМ-киназы (аtaxia telangiectasia mutated kinase) - показал, что блок клеточного цикла в G2/M-фазе при обработке МСКэ антиоксидантами обусловлен АТМ-опосредованным ответом на повреждения ДНК. Кроме того, установлено, что замедление протекания фазы синтеза ДНК под действием антиоксидантов сопровождается понижением уровня циклина А - одного из ключевых белков, регулирующих клеточный цикл на стадии синтеза ДНК.
The aim of this work was to examine the impact of sub-cytotoxic doses of antioxidants on the dynamics of the proliferative phases of the cell cycle in the synchronized cultures of human endometrial mesenchymal stem cells (eMSCs). It was shown that the addition of sub-cytotoxic doses of antioxidants to the growth medium of eMSCs progressing through the early S-phase of the cell cycle results in the dose-depended retardation of the DNA synthesis followed by the cell cycle arrest in G2/M-phase. Analysis of the accumulation of DNA strand break markers - phosphorylated histone H2AX and active ATM-kinase (ataxia telangiectasia mutated kinase) have shown that the cell cycle arrest is due to the ATM-mediated response to the DNA damage. Furthermore, it was found that retardation of the DNA synthesis, caused by the antioxidant treatment, was accompanied with the decrease of the intracellular level of the cyclin A protein - key cell cycle regulator of the S-phase progression.
