Данная работа посвящена изучению свойств белка SMC U. parvum, ортологи которого участвуют в компактизации в пространстве геномной ДНК бактерий. Предположительно, SMC U. parvum является одним из ключевых ферментов деления микоплазменных клеток. В ходе работы были применены стандартные биохимические методы и специфический анализ сдвига электрофоретической подвижности ДНК в присутствии SMC U. parvum. Дополнительно было проведено спектрофотометрическое измерение АТФ-азной активности SMC U. parvum с помощью малахитового зеленого. Белок SMC U. parvum был успешно наработан и очищен. Анализ сдвига электрофоретической подвижности ДНК позволил выявить новые ДНК-связывающие свойства SMC U. parvum. Было показано, что данный белок неспецифично связывается с линейной и кольцевой двуцепочечной ДНК (дцДНК) в отсутствии АТФ. Добавление АТФ и увеличение его концентрации приводит к усилению связывания данного белка с обоими типами дцДНК. Было охарактеризовано влияние MgCl2, KCl, pH среды на связывание белка SMC U. parvum с дцДНК. Была оценена скорость гидролиза АТФ белком SMC U. parvum в 6,2 ± 0,3 молекул в минуту. Полученные результаты востребованы в дальнейших фундаментальных исследованиях молекулярных механизмов компактизации в пространстве геномной ДНК и в разработке лекарственных препаратов против микоплазменных инфекций, нацеленных на ингибирование белка SMC.
This work is devoted to studying the properties of U. parvum SMC protein, whose orthologs are involved in the compactization of bacterial genomic DNA in space. Presumably, SMC U. parvum is one of the key enzymes of mycoplasma cell division. During the work, standard biochemical methods and the specific DNA electrophoretic mobility shift assay in the presence of SMC U. parvum were used. Additionally, the spectrophotometric measurement of the ATPase activity of SMC U. parvum was carried out using malachite green. SMC U. parvum protein was successfully produced and purified. The electrophoretic mobility shift assay revealed new DNA-binding properties of SMC U. parvum. This protein has been shown to bind nonspecifically to linear and circular double-stranded DNA (dsDNA) in the absence of ATP. The addition of ATP and an increase in its concentration leads to increased binding of this protein to both types of dsDNA. The influence of MgCl2, KCl, and pH on the binding of SMC U. parvum to dsDNA was characterized. The rate of ATP hydrolysis by SMC U. parvum was estimated to be 6,2 ± 0,3 molecules per minute. The results obtained are in demand in further fundamental studies of the molecular mechanisms of compactization of genomic DNA in space and in the development of drugs against mycoplasma infections aimed at inhibiting the SMC protein.
